For me the Raiders of the Lost Ark swordasman scene has yet to be beaten for the roar of laughter it produced with a live cinema audience.

The whole scene, totally out of the blue, Indy's facial reaction etc before he draws his gun = cinema history !

Apparently an elaborate fight scene was planned out but Harrison Ford was extremely sick and couldn't go through with it, so they came up with the idea of him just drawing his gun and shooting the guy.

Walter Matthau's scene in the Neil Simons 'California Suite' when he wakes up after a drunken sleep to find some girl completely out of it in his bed. Hes trying to wake her up before his wife gets up to his hotel room. I still convulse laughing at it. A very funny guy.
