Can someone explain to me how the notification thing works? Can you respond to them?

There are three.

there are private messages which you can respond to.

There are cool clicks which you cant respond to and wont know who clicked one for you, but you can see which of your posts that earnt one from someone though.

There are rep points which you can respond to or give one back to someone.

you have to click on the star at the bottom of their post to do that.

This is mainly used by old schoolers to comment quickly to someone Friends or foes as you can also write a note in them like a small fast pm.
No one uses neg reps.
These used to be the norm but someone got jealous cause their points wernt adding like they wanted them to cause he thought life here was a competition.

These reps,add to your punch power which is points given or taken away by positive rep or negative rep; but none takes any notice of that its just handy and good for a fast private laugh at times.
