The moon makes no difference as it doesn't take long to get there!
It might take a week off your 100 year journey to get half way across the solar system. Worth it!!!!

If you have zero gravity when you start off and if you take off from the side that never faces Earth, you are covered even more from Earths gravity so if you built a space station there, and shoot away from Earth (it isnt distance we are talking about although as you say there is a little bit saved). You would save on 95% of the fuel and size of the craft you would have make; cause it takes the most energy to get off from the Earths gravity. Initial time that would be saved as you say is minimal with distance but thats a very small part. There is a big chance we could break a few speed barriers and more time barriers, using the fuel we would have used getting off the Earth instead for more speed getting off the Moon in zero gavity.
There is no point unless you are travelling fast and far!
Any other reason is wated time! It's all about landing on random planets that look like earth! Do it or don't! I'd do it before the earth kills itself!
Take a big ship and start families on it and just breed and breed whilst travelling lifetimes through space! Take billions of rasions etc
